Es fundado en 1516 por doña Leonor de Quiñones y su hermano fray Francisco de los Ángeles Quiñones, Comisario General de la Orden franciscana.El convento se funda cumpliendo los deseos de la madre de ambos, Juana Enriquez, quien deja en su testamento todas las cosas necesarias para fundar en su Casa-Palacio, en 1515, un Convento dedicado a María Inmaculada, junto a todos los ornamentos de sucapilla y dos retablos.El nuevo convento estaría regido por la abadesa Francisca de Quiñones, hermana de doña Juana y su sobrina procedentes del convento franciscano de Palencia. Contó con los bienes aportados por la fundadora principal, y con las dotes de sus monjas y los conseguidos por donación, limosnas, rentas, bienes muebles.Siempre hubo una descendiente de los distintos Condes de Luna como profesa en el convento.El edificio sería ampliado y remodelado en varias oportunidades, gracias a los ingresos que se recibe de diferentes fuentes, lo que permite agrandar el edificio, adquiriendo las casas que con él limitan, en las cuales se construyen posteriormente la iglesia y el claustro que se encuentran a su espalda.La riqueza y fortuna del convento le permitió funcionar como casa madre, fundando otros cenobios como, el de Ponferrada, Villafranca del Bierzo y Toro.Un incendio acaecido en 1704, obligó a las religiosas a desalojar el convento y alojarse en el cercano monasterio benedictino de Santa María de Carvajal y utilizar una buena parte de las rentas conventuales, en las obligadas labores de reconstrucción.En 1808, la comunidad del Convento consta de una priora y catorce monjas. La renta anual es, entonces, de treinta y seis mil reales.La ocupación francesa no les afectó pero con posterioridad, con motivo de la Revolución en España, La Gloriosas,en 1868 la Comunidad se vio obligada de nuevo a abandonar su convento, para refugiarse en el de las Clarisas Descalzas de la ciudad, hasta 1875.Después su actividad continua con normalidad en el monasterio hasta nuestros días, que continúa activo.

It was founded in 1516 by Doña Leonor de Quiñones and her brother Fray Francisco de los Ángeles Quiñones, General Commissioner of the Franciscan Order. The building would be expanded and remodeled on several occasions, thanks to the income received from different sources, which allows the building to be enlarged, acquiring the houses that border it, in which the church and the cloister that are behind it are later built. The wealth and fortune of the convent allowed it to function as a mother house, founding other monasteries such as Ponferrada, Villafranca del Bierzo and Toro. A fire that occurred in 1704 forced the nuns to vacate the convent and stay in the nearby Benedictine monastery of Santa María de Carvajal and use a good part of the convent income, in the forced reconstruction work. In 1808, the community of the Convent consists of a pr iora and fourteen nuns. The annual income is, then, thirty-six thousand reales. The French occupation did not affect them but later, due to the Revolution in Spain, La Gloriosas, in 1868 the Community was again forced to abandon their convent, to take refuge in that of the Discalced Poor Clares in the city, until 1875. Afterwards, their activity continues normally in the monastery until today, which is still active.
