Der Zoo liegt am nördlichen Ausläufer des Duisburger Stadtwalds an der Grenze zu Mülheim an der Ruhr und tangiert den nördlichen Campus der Universität Duisburg-Essen. Die Bundesautobahn 3 teilt den Zoo in einen westlichen und einen östlichen Teil, die über eine begrünte Landschaftsbrücke verbunden sind. Die Autobahn ist für den Besucher kaum sichtbar und bietet über die Ausfahrt Duisburg-Kaiserberg eine Anbindung für mit dem PKW anreisende Besucher. Der Zoo hat eine Fläche von circa 16 Hektar."
