The church was ordered by Pope Pius IX, who in 1870 laid the first stone of the new building, but due to the annexation of Rome to the Kingdom of Italy the work was stopped. It was only thanks to the tenacity of Saint John Bosco that construction could be resumed in 1880 and finished in 1887, by the architect Francesco Vespignani.

The church is the seat of the parish of the Sacro Cuore di Gesù a Castro Pretorio, established on February 2, 1879 with the vicarial decree Postremis hisce temporibus. On February 11, 1921, Pope Benedict XV elevated it to the rank of minor basilica, with the apostolic letter Pia Societas. Subsequently, Pope Paul VI instituted the title of Cardinal of the Sacro Cuore di Gesù to Castro Pretorio, on February 5, 1965.

The façade is in the Renaissance style. Above the tower is a statue of Christ the Redeemer, placed in 1931. The interior has three naves divided by granite columns, with a transept and a dome.

The church and the annexed buildings are owned by the Salesians, who have made one of their most important centers in Rome. The founder of the church, Saint John Bosco, lived in one of its buildings from 1881 to 1884, as a commemorative plaque recalls.
