Fue un hospital social para la atención de enfermos y pobres, fundado en 1467 por la infanta doña Beatriz hija del rey Dionís de Portugal. El hospital tenía dos salas, una hombres (con ocho camas) y otra para mujeres (con cuatro camas), y estaba destinado prestar ayuda a personas cuyas enfermedades fueran curables. Además, los pacientes debían haber recibido los sacramentos para poder ser admitidos por la institución y, a diferencia de otros hospitales de la época, los enfermos no tenían un tope de días para pasar en el centro.

Las dependencias del edificio se estructuraban en torno a un claustro central, del siglo XVI, de planta rectangular, hoy totalmente cubierto por la vegetación. La iglesia original se quemó. La actual, de una sola nave, cuyos tres tramos se cubren con bóvedas de crucería con adornos plateresco, fue construida en el siglo XVI por Juan y Jerónimo Corral, en ladrillo, con espadaña a los pies y ábside de tres paños. El patio porticado consta de dos pisos con columnas de piedra. Desde el patio se accede a la iglesia.

El Ayuntamiento de Tordesillas quiere recuperar el Hospital Mater Dei y convertirlo en un centro cultural de cara a la conmemoración del V Centenario de la llegada de la Reina Juana a la villa.

Se encuentra en total abandono y en estado ruinoso.

It was a social hospital for the care of the sick and poor, founded in 1467 by the Infanta Doña Beatriz, daughter of King Dionís of Portugal. The hospital had two wards, one for men (with eight beds) and one for women (with four beds), and was intended to help people whose diseases were curable. In addition, patients had to have received the sacraments in order to be admitted by the institution and, unlike other hospitals at the time, the sick did not have a maximum number of days to spend in the center.

The dependencies of the building were structured around a central cloister, from the 16th century, with a rectangular floor plan, today completely covered by vegetation. The original church burned down. The current one, with a single nave, whose three sections are covered with ribbed vaults with Plateresque decorations, was built in the 16th century by Juan and Jerónimo Corral, in brick, with a belfry at the foot and an apse with three panels. The porticoed courtyard has two floors with stone columns. The church is accessed from the courtyard.

The Tordesillas City Council wants to recover the Mater Dei Hospital and turn it into a cultural center for the commemoration of the V Centenary of the arrival of Queen Juana to the town.

It is in total abandonment and in a dilapidated state.
