Museets opgave er at bevare bygninger og inventar, samt bruge disse til at fremvise herregårdskulturen gennem tiderne. Museet bebor herregården Gammel Estrup ved Auning og skal i det omfang det kan lade sig gøre belyse herregårdskulturen ved hjælp af husets oprindelige inventar.
Museet er et interiørmuseum, hvilket betyder, at samlingen er opstillet på en måde, som illustrerer, hvordan en herregård kunne have været indrettet til forskellige tider. Det er imidlertid umuligt at rekonstruere Gammel Estrup fra ende til anden. I forbindelse med familien Scheels salg af herregården i 1926 blev hele herregårdens indbo sat på auktion. Efter museets oprettelse i 1930 har man forsøgt at genindsamle så meget som muligt, men en total genskabelse er ikke mulig. Fortiden er væk, men med lidt omhu kan vi give folk en fornemmelse for, hvordan herregårdslivet har været i skiftende perioder.
Museet har til huse i herregården Gammel Estrups hovedbygning, hvor det har ligget siden 1930. I 1969 flyttede Dansk Landbrugsmuseum ind i herregårdens avlsbygninger fra Lyngby. De to museer ligger side om side og samarbejder om blandt andet billetsalg, men er to uafhængige institutioner. Gammel Estrup - Herregårdsmuseet er en selvejende institution og Dansk Landbrugsmuseum er et statsejet museum.

English: The permanent exhibition is an exhibition of interiors showing examples of life at the manors throughout time. The exhibition includes china, gobelin tapestries, paintings, furniture and many other articles.
The portraits of the nobilities at Gammel Estrup - the Manor Museum illustrate Danish art history with European references in a chronological time line which covers art-historic periods of renaissance, baroque, rococo and the 1800's. The historical portraits can be seen hanging authentically at Gammel Estrup - the Manor Museum. Texts which describe the personal story, the portraits and their relation to art history can be seen in the exhibition.
In the kitchen and the servants' quarters, the life of the servants is shown. The manor kitchen, the pantry, the servants' hall, a servant's room, the serving hall, the linen room, the chambermaid's room and texts and photos all illustrate life of a servant at one of the largest Danish manors at around 1918.
At present the museum is working on a full restoration of the 18th century interior on the first floor. In 2009 the restoration of the Great Hall was finished, in spring 2011 we could re-open another 5 beautiful baroque rooms, and during 2012 we were able to re-open several rococo rooms.
The museum also looks forward to welcoming visitors to The Salon from the 1790's, which opens during the summer 2013.
Every year the museum also provides the setting for different special exhibitions. On April 25 the exhibition Manors of Experiences opens. Come and experience how the Danish manors today use new ways in order to create an income which can save the manors for posterity.
