WNYO began broadcasting in 1987 as the local Fox affiliate, and was known at the time as WNYB Fox49. The station was purchased in 1988 by the same company that purchased WUTV, and the Fox affiliation was moved to that channel. WNYB was then immediately sold to a group of Christian broadcasters and began airing TBN programming until 1996.

In 1995, the owners of WNYB and a small station in Jamestown, NY worked out a deal to move the TBN facilities and call letters there, while the station in Buffalo received the new call letters WNYO. When the deal was completed September 1996, WNYO became a WB affiliate. Sinclair Broadcasting, who already owned WUTV, bought the channel in 2001 making them sister channels.

No locally produced programming originates from the station, however the local NBC affiliate, WGRZ, does broadcast a nightly 10PM newscast that originates from it's station in downtown Buffalo. WNYO did have it's own newscast for a short time in 2005-2006. Half of the odd newscast came from Sinclair Broadcasting for national news, and the other half was produced locally for hometown news. Due to low ratings, it was canceled.
