El origen del nombre de la iglesia se desconoce. Entre las posibilidades se encuentra que se le llamara así por un laurel (lauro) que hubo en tiempos cerca, una versión de dal Rio («del río»), o que en el pasado estuvo sobre una zona de marisma desecada (luprio). Se fundó en el siglo ix y se reconstruyó en 1225. El campanario data de este periodo. Hay una serie de reconstrucciones desde aquella época (incluyendo una gran renovación en 1532) y el tejado con forma de quilla data del siglo xiv. Dos de las columnas fueron botín de la Cuarta Cruzada.

The origin of the name of the church is unknown. Among the possibilities is that it was named after a laurel (lauro) that existed in nearby times, a version of dal Rio ("of the river"), or that in the past stood on an area of ??dried marsh (luprio). It was founded in the 9th century and rebuilt in 1225. The bell tower dates from this period. There are a number of reconstructions since that time (including a major renovation in 1532) and the keel-shaped roof dates from the 14th century. Two of the columns were spoils of the Fourth Crusade.
San Giacomo dall'Orio is a parish church of the Vicariate of San Polo-Santa Croce-Dorsoduro. The other churches in the parish are San Stae and San Zandegolà.
