The Palazzo presents major temporary exhibitions, some of which are based in whole or in part on the Pinault Collection, one of the largest collections of contemporary art in the world.

The architecture of Palazzo Grassi is attributed to Giorgio Massari (1687-1766) and was commissioned by the Grassi family, originally from Chioggia. It was the last palazzo to be built on the Grand Canal before the collapse of the Republic of Venice in 1797. Ownership passed to many individuals over the years and in 1983 was bought by the Fiat group which commissioned the Milanese architect Gae Aulenti to refurbish it as an exhibition space. Following the death of Gianni Agnelli, Fiat chose to terminate its involvement and in May 2005 French billionaire and art collector, François Pinault, decided to take over Palazzo Grassi.

Pinault hired Japanese architect Tadao Ando (who also restored the Punta della Dogana museum) to carry out the remodeling of Palazzo Grassi. Adopting his trademark minimal look, Ando has artfully managed to create a dialogue between the building’s original Neoclassical architecture and his modern renovation solutions.

Palazzo Grassi is also where Pinault's son Francois-Henri met actress Salma Hayek and it served as the location for their wedding vow renewal in April 2009.
