Fondée en 1843 par George Jehoshaphat Mountain, troisième évêque anglican de Québec, l’université fut d'abord un collège des arts libéraux. Sa fondation fut sanctionnée par un acte de l'Assemblée législative du Québec.
Instituée comme université par une charte royale en 1853 et affiliée à l'université d'Oxford et à l'université de Cambridge, elle porta d'abord le nom de University of Bishop's College. La charte lui conférait le pouvoir d'accorder des grades en théologie, droit, médecine, beaux-arts et musique.
En 1845, l'enseignement débuta et les premiers grades furent décernés en 1854. L'université demeura sous le contrôle de l'Église d'Angleterre jusqu'en 1947. Aujourd'hui, elle est autogérée par une corporation formée d'évêques anglicans des diocèses du Québec. Un Sénat s'occupe des affaires universitaires.
Dans la tradition des « humanités » anglo-saxonnes, l'université ne concerne que des premiers cycles.

Bishop's College was established by Bishop George Jehoshaphat Mountain on December 9, 1843, in Lennoxville, Quebec, for the education of members of the Church of England and erected into a university in 1853. The school was founded by Bishop Mountain, the third Anglican bishop of Quebec as a liberal arts college. In 1845, instruction began, and in 1854, the first degrees were granted.
It was designated as a university granting degrees in divinity, law, and medicine on January 28, 1852. An act of the Quebec Legislative Assembly ratified its foundation.
In 1853, it was established as a university by royal charter bearing the name University of Bishop's College.
