Après la guerre de 1870, les émigrants d'Alsace-Lorraine vinrent former l'essentiel de la nouvelle population juive de Châlons-en-Champagne. L'accroissement de la communauté engendrant en 1862 la création d'un rabbinat pour le département , nécessita bientôt l'oratoire de l'impasse du Moulinet et la construction d'un nouveau lieu de culte.
La nouvelle synagogue fut édifiée entre 1874 et 1875 d'après les plans de l'architecte châlonnais Alexis Vagny. L'édifice fut construit sur le terrain d'une ancienne brasserie située rue Lochet, nouvellement ouverte après la couverture de la rivière Nau effectuée en 1861.
Lors de la seconde guerre mondiale, la synagogue fut transformée en dépôt de marchandises, puis en écurie, endommageant considérablement le bâtiment; les peintures ornant les murs et les objets de culte disparurent également. Restaurée après la libération, la synagogue fut ensuite rendue au culte.
