Die ursprüngliche Marienkirche war ein romanisches Bauwerk, welches 1263 geweiht wurde. Fürst Ernst von Anhalt-Dessau legte am 25. Mai 1506 den Grundstein für die neue Kirche im gotischen Baustil. Die Weihe erfolgte am 15. Oktober 1523 unter Kardinal Erzbischof Albrecht von Mainz und Magdeburg. Als Schloss- und Stadtkirche beherbergte Sankt Marien die Grablege der askanischen Fürsten bis zum Neubau des Mausoleums im heutigen Tierpark Dessau.
Am 7. März 1945 wurde die Marienkirche durch die Bombardierung bis auf die Grundmauern zerstört. Dabei wurde auch der Prunksarg des Fürsten Leopold I. in der Fürsten- und Leopoldsgruft zerstört. In der DDR-Zeit war die Marienkirche als Mahnmal nicht wieder aufgebaut wurden. Die Restaurierung und der Wiederaufbau erfolgt, als die Kirche 1989 an den Rat der Stadt Dessau übergeben wurde.
The original St. Mary's Church was a Romanesque building, which was consecrated in 1263. Prince Ernst of Anhalt-Dessau laid on May 25, 1506 the cornerstone for the new church in the Gothic style. The consecration took place on 15 October 1523 Cardinal Archbishop Albrecht of Mainz and Magdeburg. On March 7, 1945, destroyed the St. Mary's Church by the bombing down to the ground. Here also the pomp coffin of Prince Leopold I. was destroyed in the princes and Leopold tomb. In the communist era, the St. Mary's Church was not built as a memorial again. The restoration and reconstruction takes place, when the church was handed over in 1989 to the Council of the city of Dessau.
