Les premiers colons, les Maskoutains, arrivèrent en 1757 et se mirent tout de suite à défricher les terres pour l'agriculture. Ce n'est qu'un siècle plus tard, en 1852, que Saint-Hyacinthe fut érigé en diocèse indépendant de Montréal, et ce n'est qu'en 1880 que la cathédrale actuelle fut construite.
À la fondation du diocèse, il n'y a qu'une seule paroisse dans toute la ville de Saint-Hyacinthe: celle de Notre-Dame-du-Rosaire, fondée en 1777. L'église de cette paroisse devient donc le lieu de culte de l'évêque en attendant qu'il établisse sa cathédrale. En 1853, Mgr. Jean-Charles Prince érige, dans la ville épiscopale, une nouvelle paroisse sous le nom de Saint-Hyacinthe-le-Confesseur et y fixe sa cathédrale, sans toutefois pouvoir la faire construire, faute d'argent. Quand le Séminaire de Saint-Hyacinthe prend possession de son nouvel édifice, en 1853, le vieux collège qu'il occupait depuis 1816 devient inoccupé. Mgr. Prince en prend possession et lui fait subir les transformations nécessaires pour en faire son évêché et la chapelle du collège devient alors la cathédrale. Mgr. Prince occupe ces locaux à compter du 18 janvier 1854, mais un incendie rase complètement l'immeuble et les maisons voisines le 17 mai suivant.
Pendant que Mgr. Prince trouve refuge au Séminaire, on édifie, à la hâte, une pro-cathédrale en brique rouge. Elle est ouverte au culte le 6 août 1854. En octobre de la même année, on débute la construction de l'évêché actuel sur l'emplacement du vieux collège. En 1860, aux funérailles de Mgr. Prince, la pro-cathédrale montre des signes de faiblesse sous le poids de la foule. Mais ce n'est qu'en 1875, que Mgr. Louis-Zéphirin Moreau, nouvellement nommé évêque du diocèse, songe à remplacer l'édifice. Entre temps, de 1868 jusqu'en 1876, l'église Saint-Matthieu de Beloeil lui servit de "Pro-cathédrale en exil". Le 15 août 1877, Mgr. Moreau approuve les plans et devis de l'architecte Adolphe Lévesque et signe un contrat de 50 000$. La cathédrale actuelle comme à s'ériger en 1878. L'édifice, de style roman pur, est béni par Mgr. Moreau le 4 juillet 1880. L'église , en pierre, fait 61 mètres (200 pieds) de long sur 24,4 mètres (80 pieds) de large et les deux tours, hautes et carrées, font 35 mètres (115 pieds) de haut.
Mais sa façade se révèle trop lourde pour le sol argileux sur lequel elle repose. L'édifice étant devenu dangereux, une première restauration a lieu en 1908 par les architectes Maurice Perrault et Albert Mesnard. Ces derniers consolident les fondations et les tours carrées d'origine font place à deux clochers élancés, tels que l'on peut les voir aujourd'hui. On ajoute aussi un portail et un carillon de cinq cloches est installé.
Une seconde série de réparations, en 1942, fait disparaître les balcons latéraux. Tout l'intérieur est redécoré, un nouveau baptistère est installé, tandis la crypte des évêques, sise au sous-sol, est transformée en chapelle.
À la suite du Concile Vatican II, les divers éléments du choeur sont réorganisés. L'église est également repeinte en entier et des lustres de verre de Tchécoslovaquie sont suspendus dans toute la cathédrale. En 1975, le tombeau de Mgr. Louis-Zéphirin Moreau est installé dans le transept gauche de la cathédrale, l'Église s'apprêtant à le déclarer « bienheureux ».
Les plus récentes restaurations datent de 1998 et 1999. Le parvis de cathédrale a complètement été refait. L'intérieur a été rafraîchi: une nouvelle couche de peinture fut appliquée, les boiseries furent toutes nettoyées et revernies et les dorures ont été restaurées.
La cathédrale renferme certaines oeuvres qui méritent d'être soulignées: la toile de la voûte du choeur, qui représente le Père éternel, est une oeuvre du peintre Ozias Leduc; ainsi que les tableaux du Chemin de la croix, qui datent de 1913, réalisés par le peintre parisien, Joseph Uberti.

In the foundation of the diocese, there is only a single parish in the town of Saint-Hyacinthe: that of Notre-Dame-du-Rosaire, founded in 1777. The Church of this parish thus becomes the place of worship of the Bishop until he establishes his Cathedral. While Bishop Prince found refuge in the seminary, it builds up in a hurry, a Pro-Cathedral in red brick. It is open for worship on 6 August 1854. In October of the same year, the construction of the current Bishop begin on the site of the old college. In 1860, at the funeral of Mgr. Prince, the Pro-Cathedral shows signs of weakness under the weight of the crowd. But that in 1875 as Bishop Louis-Zéphirin Moreau, newly appointed Bishop of the diocese, was thinking to replace the building. Meanwhile, from 1868 until 1876, the Church of Saint-Matthieu de Beloeil him served as "Pro-Cathedral in exile". August 15, 1877, Mgr. Moreau approves the plans and specifications of the architect Adolphe Lévesque and signed a contract of $ 50,000. The current Cathedral as to set himself up in 1878. But its facade proved too heavy for the clay soil on which it is based. The building has become dangerous, a first restoration takes place in 1908 by the architects Maurice Perrault and Albert Mesnard. They consolidate the foundations and the square towers of origin are embracing two slender bell towers, such that one can see them today. It also adds a portal and a five-Bell carillon is installed.
A second set of repairs, in 1942, dismisses the side balconies. The entire interior is redecorated, a new baptistery is installed, while the crypt of the bishops, located in the basement, is transformed into a chapel.
As a result of the Second Vatican Council, the various elements of the choir are rearranged. The Church is also repainted in its entirety and Czechoslovakia glass chandeliers are hung in the Cathedral. In 1975, the tomb of Bishop Louis-Zéphirin Moreau is installed in the left transept of the Cathedral, the Church prepares to declare it "blessed".
The most recent restorations date from 1998 and 1999. The Cathedral forecourt has been completely redone. The Interior was refreshed: a new coat of paint was applied, the woodwork were all cleaned and revernies and gold leaf has been restored.
The Cathedral contains some works that deserve to be highlighted: the vault of the choir, representing the eternal father, is a work by the painter Ozias Leduc. as well as the tables of the way of the cross, dating from 1913, made by the Parisian painter, Joseph Uberti.
