The 'Love and Rededication' ceremony is part of the city's Festival of Romance which lasts for 10 days.
Canon John Roff will start the ceremony on Sunday afternoon backed by the cathedral's singers.
During the service - which is open to all and begins at 1530 GMT - each couple will be invited to rededicate their love to one another.
Canon Roff said: "This is a wonderful opportunity for all couples to reaffirm their love for each other.
"Pre-registration is advised, but the wonderful thing about this is that couples can decide to partake on the spur of the moment too. You can't get much more romantic than that."
Chester hosted the Festival of Romance in 2004 and it attracted around 9,000 people to take part in more than 246 events.
This year's festival is expected to include a Valentine's ball and a 'hand-in-hand' charity walk around the city walls.
