The film is set in England in 1944. An OSS officer, Major John Reisman, has been tasked with taking twelve soldiers, convicted of various offences, and train them into a unit capable of infiltrating a chateau in France and capturing senior Wehrmacht officers on the eve of the D day landings. The mission is regarded as a long shot by the top brass, but Reisman gives it a go. Before being allowed to go on the mission the chosen band of men have to convince the top brass they are ready. A war game exercise takes place in England to test their ability. Using their criminal instincts, the group outwit the enemy and the adjudicators in the war game and win through.

The mission is set up and the band dispatched. At the chateau they manage to convince the Germans that there is an air raid, and that the officers should hide in the chateau's cellars. Once in the cellars the dirty dozen trap the Germans and fill the cellars with oil and grenades, which they ignite on leaving. The raid is successful, but all but two of the men are killed in the attempt.
