The community was named after William C. Greenup, a surveyor of the National Road and the first clerk of the Illinois Territorial Legislature. Greenup was plotted in 1834 by William Greenup as he surveyed the National Road from Marshall to Vandalia during the 1830's. Greenup served as the Cumberland County seat from 1843 to 1857.

Today, the town's historic district is filled with a number of antique shops. A great sense of pride exists for the town as the Main Street still runs along the original route of the National Road.
